摘要:极端嗜盐古菌利用廉价碳源生产PHBV,因具有良好的材料学性能和生产成本低廉成为目前PHA的研究热点。重点研究了解淀粉盐颗菌PHBV的提取工艺。主要研究内容:(1)优化菌体水裂解提取法的工艺。(2)优化菌体直接提取法的工艺。(3)通过对2种工艺提取的PHBV样品进行GPC、TGA、DSC分析,证明了2种工艺对PHBV样品的影响类似。菌体直接提取法提取率高于菌体水裂解提取法,但是菌体水裂解提取法得到样品的纯度较高。
PHBV production by halophilicarchaeahas been a popular study pointdue to good material properties and low production cost of this polymer. This dissertation focused on the extraction process of PHBV synthesized by Halogranumamylolyticum. The main contents were: (1)optimization of the process of cell lysis extraction method, (2)attemptionand optimization of direct cell extraction, (3)analysis of the two PHBV samples by GPC, TGA, DSC. The result showed that the two extraction processes had similar effects to PHBV characteristics. Extraction efficiency of direct cell extraction method was higher than the cell lysis extraction method. But the latter method resulted in a higher purity of PHBV.
