摘要:目的研究东亚钳蝎提取物BmKE及其醇提部位(BmKEA)对四氯化碳诱导的小鼠肝损伤的保护作用及其作用机理。方法BmKE、BmKEA、阳性对照VE灌胃第5天,小鼠腹腔注射四氯化碳造成急性肝损伤,测定小鼠血清中丙氨酸转氨酶(ALT),天冬氨酸转氨酶(AST)的活性和丙二醛(MDA)含量的变化。结果BmKE及BmKEA预处理和治疗后,肝损伤小鼠血清中ALT,AST活性均有所下降,其中,BmKEA高、中剂量组达到显著性差异(P<0.05)。BmKE低剂量组和BmKEA所有处理组血清中MDA含量极显著下降(P<0.001)。结论BmK及BmKEA通过减少肝细胞ALT和AST的泄漏,减少脂质过氧化而保护肝脏免受损伤,BmKEA是抗氧化的活性部位。
Objective To study the protective effect of extract from Buthus martensic Karch(BmKE) and the fraction separated using alcohol(BmKEA) on liver injury induced by carbon tetrachloride(CCl4) in mice and its mechanism. Methods Mice were administrated with BmKE and BmKEA with high, middle and low dosages, positive control(VitaminE) for 5d, respectively, and then acute liver injury was induced by 0.5% CCl4 ip 0.1 ml×kg^-1 The mice were killed 36h after CCl4 treatment. Alanine aminotransferase and aspartate aminotransferase( ALT and AST) activities in serum were measured. Maleic dialdehyde(MDA) in serum was detected. Results The high and middle dosages of BmKEA significantly reduced the levels of ALT and AST( P 〈 0.05 ) in serum. BmKEA remarkedly decreased the content of MDA(P 〈0. 001 ) in serum. Conclusion The BmkE and BmKEA can protect carbon tetrachloride - induced liver injury in mice by reducing the leakage of ALT and AST from the liver cells and lipid peroxidation. The BmKEA is active part of antioxidation.
