摘要:【目的】探究银杏叶提取物(GBE)和水溶性番茄浓缩物(WSTC)复合剂对高脂血症(HLP)大鼠的影响。【方法】选取40只SD大鼠,10只作为空白对照(CON)组,其余30只通过建立高脂模型后随机分为模型对照(MOD)组、阳性对照(PC)组和GBE+WSTC复合剂组。给药45 d后,测定血清中血脂指标:总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)的浓度;机理指标:低密度脂蛋白受体(LDLR)、羟甲基戊二酸单酰辅酶A还原酶(HMG-CoAR)和胆固醇7-羟化酶(CYP7A1)的质量浓度;抗氧化指标:丙二醛(MDA)、总超氧化物歧化酶(T-SOD)、谷胱甘肽(GSH)和谷胱甘肽过氧化物酶(GSH-Px)水平。【结果】与MOD组相比,在血脂指标中,GBE+WSTC复合剂组血清中TC和LDL-C的浓度极显著降低(P<0.01),TG浓度极其显著降低(P<0.001),而HDL-C浓度无显著差异(P>0.05);在机理指标中,LDLR质量浓度显著升高(P<0.05),HMG-CoAR质量浓度显著降低(P<0.05),CYP7A1质量浓度极其显著升高(P<0.001);在抗氧化指标中,MDA浓度极其显著降低(P<0.001),SOD酶活力极其显著升高(P<0.001),而GSH质量浓度和GSH-Px酶活力无显著差异(P>0.05)。【结论】GBE+WSTC复合剂组可有效改善HLP大鼠的症状,该研究为降血脂功能研究提供了理论依据。
[Objective]This study aims to investigate the effect of the Ginkgo biloba leaf extract(GBE)and water-soluble tomato concentrate(WSTC)complex on hyperlipidemia(HLP)rats.[Method]From 40 SD rats,10 rats were selected as blank control(CON)group,and the remaining 30 rats were modeled for HLP and then randomly allocated into model control(MOD),positive control(PC),and GBE+WSTC complex groups.After 45 d of administration,serum samples were collected for the measurement of the levels of lipid indicators:concentrations of total cholesterol(TC),triglycerides(TG),high-density lipoprotein cholesterol(HDL-C),and low-density lipoprotein cholesterol(LDL-C);mechanism indicators were determined:mass concentrations of low-density lipoprotein receptor(LDLR),hydroxy-3-methyl glutaryl coenzyme A reductase(HMG-CoAR),and cholesterol 7-hydroxylase(CYP7A1);antioxidant indicators were assayed:levels of malondialdehyde(MDA),total superoxide dismutase(T-SOD),glutathione(GSH),and glutathione peroxidase(GSH-Px).[Result]Compared with the MOD group,the GBE+WSTC complex group showed highly significant reducations in the concentrations of TC and LDL-C in the serum(P<0.01),and an extremely significant decrease in the concentration of TG(P<0.001),while there was no significant difference in the concentration of HDL-C(P>0.05).Among the mechanism indicators,the mass concentration of LDLR was significantly increased(P<0.05),while the mass concentration of HMG-CoA was significantly decreased(P<0.05)in the GBE+WSTC complex group in comparison with the MOD group.Particularly,the mass concentration of CYP7A1 showed an extremely significant increase(P<0.001).In terms of antioxidant indicators,the GBE+WSTC complex group showed an extremely significant declined concentration of MDA(P<0.001),an extremely significant enhanced activity of SOD(P<0.001),and no significant difference in GSH and GSH-Px(P>0.05).[Conclusion]The GBE+WSTC complex can effectively alleviate the symptoms of HLP in rats.The findings provide a theoretical basis for the study of the blood lipid-lowering function.
