摘要:报道了一种不需水解步骤测定其中虾青素及其衍生物含量的方法。其主要步骤包括:高效液相色谱分离萃取物中各酯组分,联用质谱测定各酯组分分子量;紫外-可见光分光光度法测定总酯萃取物吸光值;计算各酯组分及总酯吸光系数;计算总酯含量;计算各酯组分和总酯中虾青素结构的比率;计算萃取物中虾青素含量。由于检测结果源于紫外-可见光分光光度法测定值,且不需虾青素酯的水解操作,该方法具有操作简便、重复性好(RSD<1%)、加样回收率高(99.92%)、检测下限低(12ng/m L)等优点。
This article describes a method for the content determination of astaxanthin and its derivatives from Haematococcus plvials extract without ester hydrolysis. The main steps include : The separation and relative molecular mass determination of each ester from the extract by high performance liquid chromatography- mass spectrometry, the total ester absorbance determination by ultraviolet-visible spectrophotometry, the absorption coefficient calculations of total and individual ester, total ester content calculation, the astaxanthin ratio calculations of total and individual ester and astaxanthin content calculation. Because the result was obtained from the ultraviolet-visible spectrophotometry determination method and the ester hydrolysis was not employed, this method is easy to operate and exhibits a good repeatability (RSD 〈 1%), a nice recovery of added reference sample (99.92%) and a lower determination limit (12ng/mL) .
