摘要:目的研究灰树花多糖的复合酶解-微波萃取、超滤纯化工艺及纯化多糖的抗肿瘤活性。方法灰树花子实体经微波萃取、复合酶解(木瓜蛋白酶、纤维素酶、果胶酶的质量比为2∶2∶1)、活性炭脱色和超滤纯化得到具有不同相对分子量的灰树花多糖;采用正交试验结合响应模型,优化灰树花多糖的超滤纯化工艺;将110只荷瘤小鼠分成生理盐水组对照组和3个实验组,分别灌胃生理盐水,环磷酰胺、灰树花子实体原糖、超滤截留液和超滤透过液,研究其对荷瘤小鼠的肿瘤抑制作用。结果优化的灰树花多糖超滤工艺为:超滤温度35℃、多糖浓度20 g/L,超滤压力0.12 MPa,pH为7,超滤时间20 min;灰树花子实体原糖GFP、超滤截留液GFP-I和超滤透过液GFP-II对荷瘤小鼠的肿瘤抑制率最高分别可达40.23%、51.65%、36.24%。结论建立的响应模型能很好拟合实验结果;超滤温度和多糖浓度之间、超滤温度与pH值之间、多糖浓度与超滤压力间的交互作用极为显著;超滤时间、超滤压力对试验指标有极为显著的影响。在相同剂量下,经100kDa超滤膜分离后的超滤截留液GFP-I的肿瘤抑制率明显高于未经超滤处理多糖以及超滤滤过液多糖。
AIM To study the ultrafiltration process of Grifola Frondosa Polysaccharides(GFP) extracted by mixed enzyme-microwave and the tumor inhibiting effect of GFP.METHODS GFPs were obtained from Grifola frondosa via ultrafiltration process after being extracted by mixed enzyme-microwave extraction(mass ratio of Papain,cellulose and pectinase was 2 ∶ 2 ∶ 1 and microwave power(1 400 W)) and decolourized with activated carbon.Five-factor and four-level orthogonal experimental design was used to optimize the ultrafiltration process of Grifola Frondosa Polysaccharides.One hundred and ten tumor-bearing mice were divided into control NS group,response surface model control group,and three experimental groups raw GFP groups,the trapped liquor(GFP-Ⅰ) groups and the filtrate(GFP-Ⅱ) groups respectively to study their tumor inhibiting effects.RESULTS The optimized ultrafiltration processes were established as followings: 35 ℃ as ultrafiltration temperature,20 g/L as polysaccharide concentration,0.12 MPa as ultrafiltration pressure,20 min as ultrafiltration time and 7 as pH value.The maximum tumor inhibiting rates of raw GFP,the GFP-Ⅰ and GFP-Ⅱ groups on tumor-bearing mice were 40.23%,51.65% and 36.24%,respectively.CONCLUSION The established response model can forecast experiment results quite well.There are obvious interactions between ultrafiltration temperature and polysaccharide concentration,ultrafiltration temperature and pH,polysacaride concentration and ultrafiltration pressure.The experimental results were influenced by ultrafiltration time and pressure obviously.Tumor inhibiting rates of the GFP-Ⅰ with 100 KDa ultrafiltration membrane are higher than that of unprocessed one and the filtrate.
