摘要:目的:研究壳寡糖及其磷酸化修饰产物对小鼠脾淋巴细胞增殖及诱生细胞因子的影响。方法:采用四甲基偶氮唑盐(MTT)法检测壳寡糖及其磷酸化修饰产物在不同质量浓度条件下对小鼠脾淋巴细胞增殖的影响;用酶联免疫吸附实验(ELISA)法测定白介素2(IL-2)和γ干扰素(IFN-γ)的含量。结果:壳寡糖及其修饰产物对小鼠脾细胞均具有明显的细胞增殖作用,并存在一定的剂量依赖关系,2种实验药物均能提高IFN-γ水平;IL-2测定结果显示,壳寡糖具有明显提高脾细胞分泌IL-2的作用。结论:壳寡糖及其修饰产物具有明显的免疫增强作用,该作用与诱导小鼠脾细胞增殖、提高脾细胞中IFN-γ、IL-2的水平有关。
Objective: To investigate the effect of chitosan oligochitosan(COS-1) and the phosphorylation of chitosan oligochitosan(COS-2) on spleen lymphocyte proliferation and induction of cytokine in mice. Methods: The effects of treatments with COS-1 and with COS-2 under different quality concentrations on spleen lymphocyte proliferation were determined by MTT. The contents of IL-2 and IFN-y were determined by using ELISA. Results: COS-1 and COS-2 could increase in a significant increase of lymphocyte proliferation, and there existed certain dose-response relationship. COS-1 and COS-2 could also improve IFN-y level. Moreover, COS-2 could significantly increase the secretion of IL-2 in spleen lymphocyte. Conclusion: COS-1 and COS-2 can effectively increase lymphocyte proliferation. COS-1 and COS-2 have the ability to enhance immune responses by increasing the secretion of IL-2 and IFN-y.
