摘要:为探究鼠李糖乳杆菌R9639对小鼠肠道菌群及抗氧化能力的影响。将44只BALB/c小鼠随机分为对照组、低、中和高剂量组,分别灌胃等剂量生理盐水和不同剂量的鼠李糖乳杆菌R9639制剂,连续灌胃30 d。给药的第0、30天,采集小鼠粪便样品进行活菌计数,第30天采集小鼠粪便进行短链脂肪酸的测定,并测定小鼠小肠和肝脏组织中总超氧化物歧化酶(total superoxide dismutase,T-SOD)、丙二醛(malondialdehyde,MDA)、谷胱甘肽(glutathione,GSH)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)的水平,测定血清中白细胞介素-2(interleukin-2,IL-2)、白细胞介素-6(interleukin-6,IL-6)、白细胞介素-1β(interleukin-1β,IL-1β)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的含量。与对照组相比,灌胃鼠李糖乳杆菌R9639制剂对小鼠的体重和摄食、脏器系数、小肠的质量和长度均无明显影响;炎症因子IL-2、IL-6、IL-1β和TNF-α的水平均有所降低;小肠组织高剂量组T-SOD、GSH、GSH-Px水平均极显著升高(p<0.01),中、高剂量组MDA水平极显著降低(p<0.01),肝脏组织中、高剂量组T-SOD、GSH、GSHPx水平极显著升高(p<0.01),高剂量组MDA水平显著降低(p<0.05)。灌胃前后,与对照组和自身相比,乳杆菌和双歧杆菌含量在中、高剂量组均有极显著差异(p<0.01)。中、高剂量组的乙酸、丙酸、丁酸含量与对照组相比均有极显著差异(p<0.01)。鼠李糖乳杆菌R9639制剂对调节小鼠肠道菌群及提高抗氧化能力具有一定的作用。
This study aimed to investigated the effect of Lactobacillus rhamnosus R9639 on intestinal flora and anti-oxidation capacity in mice.Forty-four BALB/c mice were randomly divided into control,low,medium,and high dose groups.They were gavaged with equal doses of saline and different doses of Lactobacillus rhamnosus R9639 preparations for 30 days.On days 0 and 30,fecal samples of the mice were collected for viable bacteria counting and short-chain fatty acid determination.The T-SOD,MDA,GSH,and GSH-Px levels in the small intestine and liver tissues,and those of IL-2,IL-6,IL-1β,and TNF-α in the serum,were measured.Compared to the control group,there was no significant difference in body weight,food intake,organ coefficients,or weight and length of the small intestines.However,the levels of inflammatory factors IL-2,IL-6,IL-1β,and TNF-α were reduced compared to those in the control group.Compared to the control group,the T-SOD,GSH,and GSH-Px levels in the high dose group of small intestine tissues were highly significantly increased(p<0.01),and MDA levels were highly significantly decreased(p<0.01)in the medium and high dose groups.In the liver tissues,the T-SOD,GSH,and GSH-Px levels were highly significantly increased(p<0.01)in the medium and high dose groups,and MDA levels were significantly decreased(p<0.05)in the high dose group.The numbers of Lactobacillus and Bifidobacterium were significantly different(p<0.01)in the medium and high dose groups compared to those in the control group and itself pre-and post-gavage.Compared to the control group,acetic acid,propionic acid,and butyric acid in the medium and high dose groups were highly significantly different(p<0.01).Collectively,Lactobacillus rhamnosus R9639 preparation has a positive effect on regulating the intestinal flora and improving the anti-oxidation capacity of mice.
