摘要:目的:研究雨生红球藻虾青素对2型糖尿病大鼠脑组织的影响。方法:72只大鼠随机分为糖尿病模型组、正常组、雨生红球藻虾青素低剂量组【50 mg/(kg·d)】、雨生红球藻虾青素中剂量组【100 mg/(kg·d)】、雨生红球藻虾青素高剂量组【200 mg/(kg·d)】、降糖药物(盐酸二甲双胍)组,每组12只。采用腹腔注射链脲佐菌素(STZ)加高糖高脂饲料制备大鼠2型糖尿病模型。12周后,10%水合氯醛麻醉处死大鼠,4℃下取其脑部,检测GSH-Px活性、SOD活性和MDA含量;HE染色光镜下观察大鼠脑组织形态结构;免疫组化测定大鼠脑组织含IL-1β、TNF-α的细胞数量。结果:与模型组比较,高剂量虾青素可显著提高2型糖尿病大鼠脑组织GSH-Px和SOD的活性,显著降低MDA的含量,显著降低IL-1β、TNF-α的细胞数量,对糖尿病大鼠脑组织结构具有保护作用。结论:雨生红球藻虾青素对2型糖尿病大鼠脑组织具有保护作用,其作用机制可能与改善大鼠脑的氧化应激反应,降低炎性因子有关。
Objective:To study the effect of astaxanthin from Haematococcus pluvialis(astaxanthin,AST)on the brains of T2DM rats and explore the possible mechanism.Methods:randomly distributed the rats into six groups as follows:normal control group,model control group,AST low dose group,AST middle dose group,AST high dose group and positive control group(metformin hydrochloride),and established T2DM rats model with high sugar,high fat and STZ injection After 8 weeks,all the rats were killed and got brains in a low tenperature(4℃)to analyze the GSH-PX,SOD and MDA;observed pathological and morphological changes of HE dyeing brains tissue;immunohistochemistry was a method to detect the number of cells with IL-1β,TNF-αin the s rats brain.Results:Compared with model group,the GSH-PX and SOD increased evidently,and MDA obviously decreased.AST could obviously decrease cells with IL-1β,TNF-αin the diabetic rats,and had a good effect on brain tissue.Conclusions:AST has a good effect on the brains of diabetic rats,which has a good control of oxidative stress and inflammation factors and improve the brain tissue of diabetic rats.
