摘要:低聚木糖是一种效果优良的益生元,被广泛应用于各种食品中。以丰富的麦麸为原料,采用高温蒸煮耦合木聚糖酶处理制备低聚木糖具有较好的应用前景。本文以还原糖得率为指标,采用单因素实验分别研究了温度、转速、pH、酶浓度和酶解时间对木聚糖酶解效果的影响。随后,采用正交实验对温度、酶浓度和酶解时间进行组合优化,确定最佳酶解条件为:温度为65℃,转速为180 r/min,pH为5.5,酶浓度为105U/mL,酶解时间为4h。在最优条件下,还原糖得率为22.3%,低聚木糖得率为6.0%,主要由聚合度为2和3的低聚木糖组成,其比例分别为62.4%和24.9%。本文通过高温蒸煮耦合木聚糖酶对麦麸进行酶解处理制备低聚木糖具有一定的应用前景。
Xylooligosaccharide(XOS)is a prebiotic with good effect and is widely used in various foods.The preparation of XOS from wheat bran by autohydrolysis coupled with xylanase has a good prospect.In this study,the effects of temperature,rotation speed,pH,enzyme concentration and time on the yield of reducing sugar were studied by single factor experiment.Then,the temperature,enzyme concentration and time were optimized by orthogonal experiment.The optimal process conditions were obtained as follows:temperature was 65℃,rotational speed was 180 r/min,pH5.5,enzyme concentration was 105 U/mL,treatment time was 4 h.Under these optimized conditions,the yield of reducing sugars was 22.3%,and the yield of XOS was 6.0%.The XOS was mainly composed of XOS with degree of polymerization of two and three,and the proportions were 62.4%and 24.9%,respectively.In this paper,the preparation of XOS from wheat bran by autohydrolysis coupled with xylanase has a certain application prospect.
