摘要:以单位质量花生壳中白藜芦醇含量为响应值,乙醇体积分数、提取温度和提取时间为响应变量,用响应面法(RSM)确定最佳工艺条件,测定白藜芦醇粗提物对1,1-二苯基-2-三硝基苯肼(DPPH)自由基和羟自由基的清除率,用半抑制率(IC50)评价其抗氧化活性。结果表明,最佳提取条件为:乙醇体积分数62%,提取温度41℃,提取时间91 min,料液比1∶20 g/m L。在此条件下,白藜芦醇的提取量1.054 mg/g,与模型预测值1.041 mg/g基本符合。DPPH自由基和羟自由基的IC_(50)分别为(0.970±5.862)mg/m L,(0.188±2.911)mg/m L。研究表明,花生壳可以作为白藜芦醇的来源,且白藜芦醇具有较好的抗氧化活性。
Response surface methodology( RSM) was applied to obtain the optimum. Ethanol concentration,extraction temperature and time were independent variables while the yield of resveratrol was served as response. The antioxidant ability was evaluated by the scavenging rate of 1,1-diphenyl-2-trinitrophenyl hydrazine radical( DPPH) and hydroxyl radical,and half maximal inhibitory concentration( IC(50)) was used to the quantitative analysis. The results demonstrated that the maximum yield of resveratrol from peanut shell was 1. 054 mg/g at the optimum of 62% ethanol,41 ℃,91 min,and 1∶ 20 g/m L,which was in good agreement with the predicted value( 1. 041 mg/g). The IC(50) of DPPH and hydroxyl radical were( 0. 970 ± 5. 862) mg/m L and( 0. 188 ± 2. 911) mg/m L,respectively. The study showed that resveratrol has good antioxidant ability and peanut shell could be a good source of resveratrol.
