摘要:利用反相高效液相色谱/二极管阵列检测器/电喷雾质谱联用技术研究类球红细菌红色素,分离鉴定出一种组分,属于类胡萝卜素。采用C30色谱柱,在甲醇-乙腈-MTBE(3 1 1,V/V/V)为流动相,流速为1 mL·min-1,检测波长为475 nm的色谱条件下,类球红细菌红色素中有11个色谱峰在55 min内得到显著分离。借助反相高效液相色谱-电子喷雾离子源质谱联用(RP-HPLC-ESI-MS/MS)法初步确鉴定出这11个色谱峰中一种主要组分,为Philosamiaxanthin,其相对分子质量为566.5。
One main compositions of red pigment from Rhodobacter sphaeroides, which belong to carotenoids, were isolated and identified using reverse phase high performance liquid chromatography (RP-HPLC) equipped with a C30 column and a PDA detector and characterized by UV spectrometry and electrospray ionization tandem mass spectrometry/mass spectroscopy (ESI-MS/MS). As a ternary solvent system of methanol- methyl cyanide-methylt-butyl ether(MTBE) (3: 1: 1, VNN) was used as mobile phase, eleven chromatographic peaks of red pigment from Rhodobacter sphaeroides were resolved within 55 min with a C30 column of 1 mL/min flow rate at 475 nm detection. The fraction which were the main compositions of the eleven chromatographic peaks products, were identified as Philosamiaxanthin of carotenoids with the help of reverse phase high performance liquid chromatography-electrospray ionization tandem mass spectroscopy/mass spectroscopy(RP-HPLC-ESI-MS/MS), and their relative molecular weight were 566.5.
