摘要:目的探讨姜黄素(curcumin)联合有氧运动延缓D-半乳糖诱导大鼠肾脏衰老的潜在作用机制。方法 60只3月龄雄性SD大鼠随机等分为空白对照组(C组)、急性衰老组(A组)、姜黄素+急性衰老组(CA组)、有氧运动+急性衰老组(EA组)、姜黄素+有氧运动+急性衰老组(CEA组)。各衰老组连续6w腹腔注射100 mg/(kg·d) D-半乳糖建立肾脏衰老模型,并以200 mg/(kg·d)姜黄素或/和60%最大摄氧量强度有氧运动进行干预。HE/电子染色后,光镜/电镜下进行肾脏病理诊断,采用免疫组织化学法检测肾脏组织核因子E2相关因子2(nuclear factor erythroid 2-related factor 2,Nrf2)及其磷酸化和血红素氧合酶(heme oxygenase-1,HO-1)蛋白质表达,酶联免疫吸附法检测肾脏组织超氧化物歧化酶(superoxide dismutase,SOD)和γ-谷氨酸半胱氨酸合酶(γ-glutamylcysteine synthetase,γ-GCS)活性、丙二醛(malondialdehyde,MDA)含量,荧光比色法测定脂褐质(lipofuscin,LPF)含量。结果与A组比较,CA、EA、CEA组均可有效激活Nrf2信号通路,上调下游抗氧化酶活性,缓解氧化应激,延缓肾脏衰老;且姜黄素联合有氧运动激活效果增强。结论姜黄素联合有氧运动能够调控Nrf2信号通路,缓解氧化应激,延缓大鼠肾脏衰老。[营养学报,2021,43(1):79-85]
Objective To explore the actions of curcumin combined with aerobic exercise in delaying D-galactose induced renal aging in rats. Methods Sixty 3-month-old male SD rats were randomly divided into 5 groups(n=12/group): control group(group C), acute aging group(group A), curcumin + acute aging group(group CA), aerobic exercise + acute aging group(group EA), and curcumin + aerobic exercise + acute aging group(group CEA). Rats from aging groups were injected intraperitoneally with 100 mg/(kg·d) D-galactose for 6 consecutive weeks to establish the renal aging model. Intervention for group CA, EA and CEA was performed with curcumin at 200 mg/(kg·d) or/and aerobic exercise at 60% maximum oxygen uptake intensity. Histological changes in the kidney was examined under light microscope/electron microscope after H&E staining. The expression of renal nuclear factor erythroid 2-related factor 2(Nrf2), phosphorylated Nrf2 and heme oxygenase-1(HO-1) were detected by immunohistochemical method. Renal superoxide dismutase(SOD) andγ-glutamylcysteine synthetase(γ-GCS) activity, malondialdehyde(MDA) concentration were detected by enzyme-linked immunosorbent assay. Renal lipofuscin(LPF) concentration was detected by fluorescence colorimetry. Results Compared with group A, Nrf2 signaling pathway was activated in group CA, EA and CEA, the activity of downstream antioxidant enzyme was up-regulated. Alleviated oxidative stress may delay renal aging. The activation effect of Nrf2 was enhanced.by curcumin combined with aerobic exercise. Conclusion Curcumin combined with aerobic exercise activates the Nrf2 signaling pathway, relieves oxidative stress, and may delay the renal aging in rats. [ACTA NUTRIMENTA SINICA, 2021, 43(1):79-85]
