摘要:建立了一种简单快速的检测饮料中咖啡因的超高效液相色谱方法。饮料经简单的过滤,然后采用ACQUITYUPLC@BEHC18色谱柱进行分离,以甲醇.水溶液为流动相进行梯度洗脱,流速为0.4mL/min,柱温为35℃,检测波长为272.2nm。咖啡因能与样品中的成分达到基线分离,咖啡因的检测限为0.4mg/L;咖啡因浓度在5-150mg/L时,峰面积与浓度成良好的线性关系,r=0.9999;日内(n=6)测定的相对标准偏差(RSD)为0.43%,日间(n=4)测定的RSD为t.04%;样品的加标回收率在99.81%-101.35%之间。该方法简单、快速、准确,适用于饮料中咖啡因的测定,可以为保证饮料的品质和维护人民健康提供技术保证。
A simple and fast ultra-performance liquid chromatography (UPLC) method was established to determine the content of caffeine in beverages. After filtration, UPLC determination for caffeine in beverages was performed in an ACQUITY UPLC~ BEH C18 column and detected at 272.7nm. A gradient separation was achieved using with methanol-ultrapure water as mobile phase. Flow rate of the mobile phase was set at 0.4mL/min and the column temperature was 35"C. Caffeine and other ingredients in the samples were separated well. The detection limit of caffeine was found to be 0.4mg/L. The concentration of caffeine and the peak area had good linearity in the range of 5--150mg/L, with the correlation coefficient of 0.9999. The relative standard deviations of intra-day (n=6) and inter-day (n=4) determination were 0.43% and 1.04%, respectively. The standard recovery rate was between 99.81% and 101.35%. The method has been successfully applied to determine caffeine in beverages. It was simple, rapid and accurate, which can provide technical support to ensure the quality of beverage and maintain people's health.
