摘要:目的:探讨姜黄素(Curcumin)对大气细颗粒物PM_(2.5)(particulate matter 2.5μm)滴注致大鼠急性肺损伤的影响。方法:将50只SD大鼠随机分为5组:空白对照组、PM_(2.5)模型对照组、低剂量姜黄素(100 mg·kg^(-1))+PM_(2.5)组、中剂量姜黄素(200 mg·kg^(-1))+PM_(2.5)组、高剂量姜黄素(300 mg·kg^(-1))+PM_(2.5)组。各剂量姜黄素组用姜黄素连续灌胃14 d,空白对照组、模型对照组用生理盐水灌胃。第15 d灌胃后,除空白对照组外的各组大鼠气管滴注PM_(2.5)(9.0 mg·kg^(-1))混悬液染毒,24 h/次共3次,空白组气管滴注生理盐水。末次染毒24 h后,收集血样、支气管肺泡灌洗液(Bronchoalveolar lavage fluid,BALF)、肺组织匀浆和病理切片。分析测定血清中丙二醛(Malondialdehyde,MDA)、超氧化物歧化酶(Superoxide dismutase,SOD)、BALF中的乳酸脱氢酶(Lactate dehydrogenase,LDH)、酸性磷酸酶(Acid phosphatase,ACP)、碱性磷酸酶(Alkaline phosphatase,AKP)水平;肺匀浆液中肿瘤坏死因子α(Tumor necrosis factor-α,TNF-α)、白细胞介素-1β(Interleukin-1β,IL-1β)水平。结果:与空白对照组比较,PM_(2.5)模型对照组大鼠的MDA、AKP、ACP、LDH、TNF-α、IL-1β水平增加、SOD活力降低(P<0.05或P<0.01);与PM_(2.5)模型组比较,低剂量姜黄素+PM_(2.5)组能降低ACP、LDH、TNF-α、IL-1β水平,提升SOD活力(P<0.05或P<0.01),MDA、AKP水平无显著差异;中、高剂量姜黄素+PM_(2.5)组能降低MDA、AKP、ACP、LDH、TNF-α、IL-1β水平,升高SOD活力(P<0.05或P<0.01),中、高剂量姜黄素组能改善大鼠肺组织病理变化。结论:姜黄素对PM_(2.5)滴注致大鼠急性肺部氧化应激及炎症损伤具有保护作用。
To investigate the effect of curcumin on acute lung injury in rats induced by atmospheric PM_(2.5) instillation,50 SD rats were randomly divided into 5 groups:blank control group,PM_(2.5) model control group,low-dose curcumin(100 mg·kg^(-1))+PM_(2.5) group,medium dose curcumin(200 mg·kg^(-1))+PM_(2.5) group,high-dose curcumin(300 mg·kg^(-1))+PM_(2.5) group.Each curcumin dose group was continuously perfused with curcumin for 14 days.The blank control group and the model control group were given normal saline by gavage.After intragastric administration on the 15th day,except for the blank group being dosed with saline by tracheal drip,the rats in all other groups were infected with PM_(2.5)(9.0 mg·kg^(-1))suspension by tracheal infusion for three times at 24 hour intervals.24 hours after the last exposure,blood samples,bronchoalveolar lavage fluid,lung tissue homogenates and pathological sections were collected.The levels of malondialdehyde(MDA),superoxide dismutase(SOD),lactate dehydrogenase(LDH),acid phosphatase(ACP)and alkaline phosphatase(AKP)in BALF were analyzed and measured in serum.The levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in the lung homogenate were measured.Results showed that compared with the blank control group,the levels of MDA,AKP,ACP,LDH,TNF-αand IL-1βincreased and SOD activity decreased in the PM_(2.5) model control group(P<0.05 or P<0.01);compared with the PM_(2.5) model group,the levels of ACP,LDH,TNF-αand IL-1βdecreased in the low-dose curcumin+PM_(2.5) group and SOD activity increased(P<0.01).Compared with the PM_(2.5) model group,the low-dose curcumin+PM_(2.5) group could reduce the levels of ACP,LDH,TNF-α,IL-1βand elevate the SOD activity(P<0.05 or P<0.01),and there was no significant difference in the levels of MDA and AKP;the medium-dose and high-dose curcumin+PM_(2.5) groups could reduce the levels of MDA,AKP,ACP,LDH,TNF-α,IL-1βand elevate the SOD activity(P<0.05 or P<0.01),and the medium-dose and high-dose groups could improve the histopathological changes of rat lung.It can be concluded that curcumin has a protective effect on acute lung oxidative stress and inflammatory injury caused by PM_(2.5) drip injection in rats.
